Developing LC methods for optimum performance of basic analytes can be challenging. The familiar struggles with adequate retention and tailing peak shape which impacts resolution and the ability to identify impurities as well as achieve baseline separation can be overcome with knowledge of the options that are available. This presentation will detail method parameters such as pH, stationary phase, mobile phase additives, choice of detector, and how they can be tailored for better performance for basic analytes and their impact on acids and neutrals. Examples will include separations of small molecule pharmaceuticals, drugs of abuse, and peptides to demonstrate how improvements can be achieved on these common basic analyte chromatography challenges.
